Literature search strategy - Identifying the key publications for transcriptomic biomarkers in genetic toxicity

Discrimination between genotoxic and non-genotoxic carcinogens is critical to chemical risk assessment. However, traditional genotoxicity assessment is based on a battery of assays that can produce discordant results and equivocal conclusions. The development of new strategies employing toxicogenomic methods is desirable.  The Steering Committee of the 8th International Workshops on Genotoxicity Testing (IWGT) convened an expert working group (EWG) to critically assess previously reported transcriptomic biomarkers of genotoxicity. References for a systematic review of these biomarkers were identified using an initial collection of 18 reports selected by the EWG. This initial collection was expanded using the PubMed functionality that identified and retrieved an additional set of 1323 potentially relevant references as most similar to the expert-driven initial collection of “seed” references.” References from this expanded collection were filtered, screened, and evaluated for risk of bias to identify the most relevant studies meeting defined quality criteria. The EWG will evaluate the predictive performance of the biomarkers reported in these studies for discrimination between genotoxic and non-genotoxic modes of action and their applicability for risk assessment of chemicals, consumer products, and pharmaceuticals.    Disclaimer: The views expressed in this document are solely those of the author and do not necessarily reflect those of the Agency. Impact/Purpose

Discrimination between genotoxic and non-genotoxic carcinogens is critical to chemical risk assessment. The Steering Committee of the 8th International Workshops on Genotoxicity Testing (IWGT) convened an expert working group (EWG) to critically assess previously reported transcriptomic biomarkers of genotoxicity. Here we present the approach used for identification, screening, and evaluating the quality of studies reporting on biomarkers of genetic toxicity. The biomarkers will be evaluated for their predictive performance and their applicability for risk assessment of chemicals, consumer products, and pharmaceuticals.
