Presentation on Case Studies: Application of APROBA to the Acrolein and Chloroform

This sub-product consists of a presentation on two cases studies on the derivation of probabilistic reference values. In the first case study, the unified probabilistic framework was applied to incidence of lesions in the nasal epithelium in male rats exposed to acrolein, using the software tool Approximate Probabilistic Analysis (APROBA). The probability distribution was calculated for the risk-specific dose, that is, the dose at which 1% of the human population is estimated to experience minimal lesions, and a probabilistic reference value was computed as the 5th percentile of this distribution. A deterministic reference value was also derived for comparison, and a sensitivity analysis of the probabilistic reference value was conducted investigating alternative assumptions for the point of departure type and exposure duration.
In the second case study, APROBA was applied to a number of effects from exposure to chloroform by inhalation. Similar to acrolein, probability distributions were calculated for the risk-specific doses for these effects, and a probabilistic reference value was computed. Also, deterministic reference values were derived, and a sensitivity analysis of the probabilistic reference value was conducted.
