EPA announced the availability of this final report as supporting material for the Federal Register Notice: Drinking Water: Perchlorate Supplemental Request for Comments as announced in an August 19, 2009 Federal Register Notice. EPA originally released this document in draft form as supporting material for the Preliminary Regulatory Determination on Perchlorate (FR Notice) in October 2008. The document has since been revised in response to comments from independent external peer review. A copy of the PBPK model code is available. In addition, the final peer review report and a summary of external peer review comments and EPA responses are available from the draft report web page.
This report provides an analysis of perchlorate-mediated inhibition of the sodium-iodide symporter (NIS) in humans using published PBPK models, focusing on the degree of NIS inhibition as a function of lifestage. The models provide information that may be used to address differences in human responses to perchlorate across lifestages.

History/Chronology

Date Description
01- Feb 2005 EPA established a reference dose for perchlorate, which was consistent with the reference dose recommended by the National Research Council's 2005 report. IRIS Risk information about perchlorate and perchlorate salts.
02- Jul 2008 EPA made a final regulatory determination for priority contaminants on the second contaminant candidate list.
03- Oct 2008 EPA made a preliminary regulatory determination for Perchlorate and released the draft report, Inhibition of the Sodium-Iodide Symporter By Perchlorate: An Evaluation of Lifestage Sensitivity Using Physiologically Based Pharmacokinetic (PBPK) Modeling (External Review Draft), for public review and comment.
05- Dec 2008 Following an independent external peer review, EPA released the consolidated comments received on the draft report. At the same time, EPA also issued an Interim Drinking Water Health Advisory for Perchlorate PDF (49pp, 406K, About PDF).
06- Aug 2009 EPA released the summary of external peer review comments and disposition for the 2008 external review draft report.
07- Aug 2009 EPA publishes the final report.
